چکیده

Atherosclerosis is a complex disease, the onset of which depends on various components of the vascular system, metabolism and immune system. It generates the fibro-fatty plaque or stable plaque characterized by accumulation of lipids in the intima of the arteries, a fibrous cap covering the atheromacore which may consists in lipid-laden cells (macrophages andvascular smooth muscle cells), proteoglycans, collagen, elastin, foam cells and, sometimes, cholesterol crystals needle-like clefts, fibrinand neovessels. The progression of the plaque leads to unstable atherosclerotic lesions. The result will be a large plaque, consisting of an evident lipid core surrounded by a fibrous cap, infiltrates of immunocompetent cells and calcium deposits. Advanced plaque is characterized by macrophages invasion and by the thinning of the fibrous cap. Progression of atherosclerotic plaque can lead to its rupture and result in the occlusion of an artery or in the formation of a thrombus. Apoptosisinthe fibrouscap, rich in vascular smooth muscle cells and macrophages, and its subsequent weakening seems to beanimportantregulatorofplaque stability. In our study, we collected specimens from stable atherosclerotic plaques in the right or left internal carotid artery of patients with clinical symptoms. Histology and histochemistry were performed in specimens for cell identification and detection of structural alterations. Immunohistochemical analysis related to caspase-3 and N-cadherin was performed in order to highlight the pro-survival role of N-cadherin against apoptosis in the stable atherosclerotic plaques. Our results showed that when expression of N-cadherin is evident and strong in the stable atherosclerotic plaques, apoptosis, expressed by caspase-3 immunostaining, is not detected, as reported by recent literature. The aim of our study was to acquire greater knowledge on the biological mechanisms related to plaque vulnerability in order to develop new therapies to maintain atherosclerotic plaque stability avoiding its rupture which could determine consequences such as thrombosis.

DETECTION AND RESTRICTION ANALYSIS OF C YTOMEGALOVIRUS DNA PERSISTING IN HUMAN ATHEROSCLEROTIC PLAQUES USING POLYMERASE CHAIN REACTION

The polymerase chain reaction (PCR) as applied to detection of a foreign DNA in clinical specimens could provide a sensitive instrument for rapid detection of viral DNA persisting in tissues of patients suspected of latent infection. Human cytomegalovirus (HCMV) DNA was found in arterial plaques of patients with atherosclerotic lesions using a PCR assay with nested primer oligonucleotides ...

Effect of Helicobacter pylori DNA in human atherosclerotic plaques

Introduction: A number of studies have demonstrated that infectious mico organisms like helicobacter pylori may play a role in the process of atherosclerosis. We, here, aimed to investigate the effect of Helicobacter pylori DNA in atherosclerotic plaques in patients with coronary artery disease. Methods: In a cross-sectional study, 85 patients undergoing coronary artery bypass graft (CAB...

Decreased cathepsin K levels in human atherosclerotic plaques are associated with plaque instability

Investigating the determinants and dynamics of atherosclerotic plaque instability is a key area of current cardiovascular research. Extracellular matrix degradation from excessive proteolysis induced by enzymes such as cathepsin K (Cat K) is implicated in the pathogenesis of unstable plaques. The current study assessed the expression of Cat K in human unstable atherosclerotic plaques. Specimens...

Evidence for increased collagenolysis by interstitial collagenases-1 and -3 in vulnerable human atheromatous plaques.

BACKGROUND Several recent studies attempted to classify plaques as those prone to cause clinical manifestations (vulnerable, atheromatous plaques) or those less frequently associated with acute thrombotic complication (stable, fibrous plaques). Defining the cellular and molecular mechanisms that underlie these morphological features remains a challenge. Because interstitial forms of collagen de...

Detection of Helicobacter pylori in human carotid atherosclerotic plaques.

BACKGROUND AND PURPOSE Several lines of evidence point toward a relationship between infection and atherosclerotic vascular disease. Thus, infection and inflammation often precede ischemic neurological events. Transient alterations in coagulation and direct arterial invasion by certain microorganisms have been reported. Helicobacter pylori infection is the major cause of peptic ulcer disease an...
